Product Name 4x Multiplex PCR Master Mix, Low-Background Version (1-2000-Plex Pathogen tNGS)
Catalog No. REKI-0107
Description A 4x concentrated master mix developed for two-round multiplex amplification in amplicon-based library construction. Combined hot-start chemistry and a tuned buffer system scale from singleplex up to 2000-plex panels while keeping primer-dimer and background signatures low. Amplicons across a 25 to 75 percent GC window amplify with high uniformity, specificity and sensitivity, and 30 to 500 primer pairs of differing amplicon sizes work in the same reaction, with headroom into the several-thousand-plex range for amplicon capture. The mix accepts reverse-transcribed cDNA directly, so RNA viruses join DNA pathogens in a single workflow without an intermediate cleanup.
Intended Use Pathogen targeted NGS panel construction, tNGS amplicon library preparation, co-detection of DNA and RNA pathogens in syndromic panels.
Principle / Technology Two rounds of hot-start multiplex PCR build and enrich amplicon libraries; target-specific primers define the panel while the master mix supplies enzyme, buffer and dNTPs.
Performance Range / Specifications Panel depth 1 to 2000-plex; 30 to 500 amplicon pairs per reaction; amplicon GC coverage 25 to 75 percent; high uniformity, specificity and sensitivity.
Specificity Reverse-transcribed samples enter multiplex PCR directly without purification.
Reaction Conditions / Protocol Combine template, panel primers and 4x master mix, run the two-round multiplex programme, then proceed to library finishing.
Components / Formulation Hot-start multiplex polymerase in 4x premix format, optimised reaction buffer, dNTPs.
Storage Conditions -25 to -15 °C
Shelf Life 2 years
