Product Name Recombinase Polymerase Amplification (RPA) Lyophilized Reaction Pellet Kit
Catalog No. NATR-HMM-0144
Description A lyophilized reagent kit for isothermal nucleic acid amplification using the recombinase polymerase amplification method. Each reaction pellet contains all essential enzymes and cofactors pre-dispensed in a dry, room-temperature-stable format requiring only the addition of primers, template, and magnesium acetate for initiation. RPA operates at a constant low temperature, eliminating the need for thermal cycling equipment. This format is suited for point-of-need and field-deployable molecular testing workflows.
Intended Use Isothermal amplification of DNA targets at constant low temperature for rapid nucleic acid detection outside of laboratory settings.
Principle / Technology Recombinase polymerase amplification at constant 37–42 °C
Detection Method Real-time fluorescence, lateral flow dipstick, or gel electrophoresis
Sample Type Purified DNA from clinical specimens, environmental samples, food matrices, or cultured organisms
Performance Range / Specifications Amplification time: 10–20 min to detectable signal; compatible with 80–300 bp amplicons
Sensitivity / LOD Detection limit: 1–10 copies of target DNA per reaction
Specificity Primer-dependent target specificity; T4 UvsX recombinase and Bsu DNA polymerase function optimally at 39 °C
Reaction Conditions / Protocol Rehydrate pellet with 29.5 µL rehydration buffer plus primers; add 2.5 µL magnesium acetate to initiate; incubate 20 min at 39 °C
Components / Formulation Lyophilized RPA pellets (8-strip tube format), rehydration buffer, magnesium acetate solution (280 mM), positive control template and primers
Storage Conditions Lyophilized pellets: room temperature for up to 3 weeks or 2–8 °C for long-term; rehydration buffer and magnesium acetate: –20 °C
Shelf Life 12 months at 2–8 °C in sealed pouch with desiccant
Package Specifications 96 reactions (12 × 8-strip tubes)
Product Form Lyophilized pellet in 0.2 mL PCR tube strips
Quality Control Each pellet lot validated with control template to confirm amplification within 15 min; NTC contamination check negative
Key Features Room-temperature shipping and short-term storage removes cold chain dependency for field deployment
Purity DNase/RNase free; PCR inhibitor free; A260/A280 ≥1.8 for nucleic acid products

Activity / Unit Definition Polymerase activity defined as nmol dNTP incorporated per 30 min per unit; other activities per product specification
Molecular Weight As specified per DNA markers or protein components
Source / Origin Recombinant enzymes expressed in E. coli; synthetic oligonucleotides; ultrapure reagents
pH Range / Optimal pH pH 7.5–8.5 for PCR; pH 8.0 for most molecular biology applications
Shipping Conditions Cold packs or dry ice for enzymes; ambient for buffers and DNA markers
Expiration Date / Stability 12–24 months at –20 °C for enzymes; 12–24 months at room temperature for buffers
Regulatory / Compliance Research use; ISO 13485 manufacturing for select products
Compatibility Compatible with standard thermal cyclers, real-time PCR instruments, and gel electrophoresis systems
Recommended Buffer System Tris-HCl, KCl, MgCl₂-based reaction buffer; TE for nucleic acid storage
Application Notes / Precautions Use dedicated PCR workspace; aliquot enzymes to avoid contamination; monitor no-template controls
Batch-to-Batch Consistency Enzyme activity within ±15% of reference lot; PCR performance verified with control template
