Description Alta DiagnoTech RT-LAMP probe master mix is a ready-to-reconstitute in-tube lyophilized reagent for DNA or RNA isothermal amplification.
Intended Use For DNA or RNA isothermal amplification.
Principle / Technology A dried formulation supplies the core buffer and enzyme system; users add primer/probe mix and template before isothermal amplification.
Detection Method Probe-based fluorescence detection
Reaction Conditions / Protocol Use one dried dose with 5 µL 10× primer/probe mix and 45 µL RNA template. The 10× primer/probe mix contains 16 µM FIP/BIP, 2 µM F3/B3, 4 µM Loop F/B and 4 µM probe. Run 30 cycles at 65°C for 60 seconds with FAM fluorescence acquisition. The reaction temperature may be optimized from 62°C to 68°C.
Components / Formulation Reaction buffer containing Mg2+ and dNTPs, Bst DNA polymerase and thermostable reverse transcriptase enzyme mix, lyophilization protectants. The 10× primer/probe mix contains 16 µM FIP/BIP, 2 µM F3/B3, 4 µM Loop F/B and 4 µM probe.
Storage Conditions Store at 2-8°C.

Package Specifications Available in 96-, 960- and 9,600-test formats, configured as 8 tests per strip with 12, 120 or 1,200 strips.

Compatibility Use nucleic-acid template dissolved in DEPC-treated water. Reaction temperature may be optimized for the selected primer set.
Application Notes / Precautions Use a controlled workflow for reaction setup, sample handling and loading. Prepare the reaction mix in a clean bench and add template in a separate room or ventilated enclosure to reduce false-positive contamination.
