Description A rapid amplification of cDNA ends system that combines template-switching reverse transcription with high-fidelity PCR to recover unknown transcript termini.

Principle / Technology A template-switching reverse transcriptase generates full-length cDNA, followed by high-fidelity amplification of the 5′ or 3′ end without a separate adapter-ligation step.
Detection Method PCR amplification and downstream gel analysis or sequencing.

Performance Range / Specifications 10 reactions; reverse transcription demonstrated with transcripts up to 15 kb and abundance as low as 0.9 RPKM; tested transcript GC range approximately 36–70%.
Reaction Conditions / Protocol Prepare full-length cDNA with the template-switching reverse-transcription module, then amplify the target 5′ or 3′ end with a gene-specific primer and the supplied PCR system.

Application Notes / Precautions For low-abundance targets, RNA input can be increased up to 4 µg. Use a gene-specific primer with a melting temperature of at least 65°C. For amplicons up to 3 kb, allow approximately 3 minutes of extension and add about 30 seconds per additional kilobase.
