Description A purple universal SYBR qPCR master mix using a dUTP and heat-labile UDG system to remove carryover products before amplification.
Intended Use Relative and absolute nucleic acid quantification by dye-based real-time PCR.
Principle / Technology Heat-labile UDG removes uracil-containing carryover DNA at room temperature and is inactivated during initial heating, while a hot-start polymerase drives specific qPCR amplification.
Detection Method Intercalating-dye real-time PCR.
Sample Type DNA or cDNA templates.
Performance Range / Specifications 500 or 2,500 reactions; carryover removal above 99.99% was demonstrated under the evaluated conditions.
Specificity Antibody-mediated hot-start blockade remains effective up to approximately 55°C, helping suppress nonspecific amplification.
Reaction Conditions / Protocol Add primers and template to the master mix. The carryover-control enzyme acts at room temperature and is inactivated during heating at approximately 50–55°C before qPCR amplification.
Components / Formulation qPCR master mix containing hot-start DNA polymerase, intercalating dye, dUTP, heat-labile UDG, universal passive-reference dye and purple tracking dye.
