Description A routine plasmid miniprep kit using optimized alkaline lysis and selective spin-column purification. The process does not require phenol/chloroform extraction, alcohol precipitation or column equilibration.
Intended Use Plasmid DNA purification for restriction digestion, PCR, sequencing, ligation and transformation.
Principle / Technology SDS-alkaline lysis followed by selective silica-membrane binding, washing and elution.
Detection Method Downstream plasmid DNA analysis.
Sample Type 1-5 mL overnight bacterial culture.
Performance Range / Specifications Approximately 30-minute workflow; column binding capacity up to 35 µg plasmid DNA; compatible with plasmids demonstrated from approximately 3-10 kb; 100 reactions.
Specificity Selective plasmid DNA recovery with removal of protein, genomic DNA, RNA and other bacterial impurities.
Reaction Conditions / Protocol Harvest the culture, resuspend, lyse and neutralize, clarify the lysate, bind plasmid DNA to the spin column, wash and elute.
Components / Formulation Resuspension, lysis, neutralization, wash and elution reagents with RNase, silica spin columns and collection tubes.
Storage Conditions Store at 15-25°C. After RNase is added to the resuspension buffer, store at 2-8°C for up to 6 months.
