Description A non-viral stable gene transfer system consisting of a transposon donor plasmid and a transposase expression helper plasmid for the efficient, stable genomic integration of large transgene cassettes in mammalian cells.
Intended Use Generation of stable cell lines through transposon-mediated genomic integration of large transgenes (up to 100 kb); suitable for multi-gene expression systems, inducible expression constructs, and cell line engineering without viral vector biosafety concerns.
Principle / Technology PiggyBac transposase recognizes terminal inverted repeat sequences flanking the transgene cargo on the donor plasmid, excises the cargo, and catalyzes its integration at TTAA tetranucleotide sites in the host genome with precise footprint-free excision capability.
Detection Method Stable integration verified by antibiotic selection and colony counting, transgene copy number by qPCR, integration site analysis by splinkerette PCR
