Product Name DEAE-Dextran Transfection Reagent, High Molecular Weight
Catalog No. ATR-CTR-0015
Description A high-molecular-weight diethylaminoethyl-dextran polymer solution for the transient transfection of mammalian cells through a simple and economical chemical method, historically significant and still used for specific applications.
Intended Use Transient transfection of COS cells and other cell lines for recombinant protein production; transient expression in BHK, CV-1, and other cell lines requiring high plasmid copy number per cell.
Principle / Technology Positively charged DEAE-dextran polymer complexes negatively charged DNA and binds to the cell surface via electrostatic interactions; treatment with chloroquine or DMSO shock enhances endosomal release and DNA delivery to the nucleus.
Detection Method Reporter gene expression by enzymatic assay or flow cytometry, transient protein production measurement
Sample Type COS-1, COS-7, CV-1, BHK-21, and other cell lines susceptible to DEAE-dextran transfection
Performance Range / Specifications Transient expression in 30–80% of cells depending on cell line and protocol; high plasmid copy number per cell achieved
Sensitivity / LOD N/A
Specificity N/A
Reaction Conditions / Protocol Mix DNA with DEAE-dextran in serum-free medium; incubate with cells for 30 min to 4 hours; remove DNA-DEAE mixture; apply DMSO or chloroquine shock (optional); add complete medium; assay 48–72 hours post-transfection.
Components / Formulation DEAE-dextran hydrochloride (average MW 500,000), lyophilized or in sterile water at 10 mg/mL
Storage Conditions 15–30°C (dry powder); 2–8°C (solution); protect from moisture
Shelf Life 36 months (dry); 12 months (solution)
Package Specifications 1 g (powder), 10 mL (10 mg/mL sterile solution)
Product Form Lyophilized powder or sterile-filtered liquid
Quality Control Molecular weight by GPC, transfection validation with COS-7 reference cells, endotoxin <0.1 EU/mg, sterility
Key Features Cost effective; high plasmid copy number; established protocol; suitable for COS cell expression; economical for large-scale transient production

For research use only, not for clinical use.
