| Product Name | MTT Cell Proliferation and Cytotoxicity Assay Kit |
| Catalog No. | RESK-0155 |
| Description | MTT (methylthiazolyldiphenyl-tetrazolium bromide), also known as thiazolyl blue tetrazolium bromide, is a yellow dye that has widely replaced traditional trypan blue staining and radioisotope incorporation methods for measuring cell viability, cell proliferation and cytotoxicity. This kit uses high-purity MTT as the indicator and, based on the classic procedure, employs a unique formazan dissolving solution formulation that directly dissolves the formazan precipitate without removing the original culture medium, thereby avoiding the operational error caused by partial loss of formazan during medium removal. The kit features low background, high sensitivity, a wide linear range and simple operation. Matching disposable syringe-driven filters and syringes are also provided for added convenience. |
| Intended Use | Detection of cell viability, cell proliferation and cytotoxicity in cultured cells. |
| Principle / Technology | MTT is a positively charged, cell membrane-permeable compound dye that accepts hydrogen ions. Succinate dehydrogenase in the mitochondria of living cells reduces exogenous MTT to water-insoluble, dark blue MTT-formazan crystals; dead cells lack this capability. The formazan crystals cannot cross the cell membrane and accumulate in proliferating, undamaged cells. The kit's unique formazan dissolving solution directly dissolves the crystals without removing the culture medium, and the absorbance at 570 nm reflects the amount of formazan formed, which is directly proportional to the number of viable cells. |
| Detection Method | Colorimetric readout: absorbance measured at 570 nm with a microplate reader. |
| Sample Type | Cultured proliferating cells. |
| Performance Range / Specifications | Low background, high sensitivity, wide linear range and simple operation. The unique formazan dissolving solution directly dissolves the formazan precipitate without removing the original culture medium, avoiding operational errors caused by partial loss of formazan. |
| Sensitivity / LOD | High sensitivity (see Performance Range / Specifications). |
| Specificity | MTT is reduced specifically by mitochondrial succinate dehydrogenase of metabolically active, living cells; dead cells do not reduce MTT. The amount of formazan generated is directly proportional to the number of viable cells. |
| Reaction Conditions / Protocol | 1. Add the MTT working solution to the cell culture and incubate to allow living cells to reduce MTT to dark blue formazan crystals. 2. Add the formazan dissolving solution directly to dissolve the formazan precipitate without removing the original culture medium. 3. Measure the absorbance at 570 nm on a microplate reader; the signal is proportional to the number of viable cells. |

| Regulatory / Compliance | For research use only. |
| Application Notes / Precautions | 1. Prepared MTT working solution is yellow and must be protected from light; prolonged exposure to light causes inactivation. Do not use the solution if it turns gray-green. 2. MTT solution solidifies at low temperatures such as 4 °C or in an ice bath; warm it briefly in a 20-25 °C water bath until fully dissolved before use. 3. The prepared MTT solution does not need to be filtered. |
For research use only, not for clinical use.
