Product Name ATP Luminescent 3D Cell Viability Assay Kit
Catalog No. RESK-0159
Description ATP is the most important energy molecule in cells. It reflects the level of cellular metabolism and shows a good linear correlation with the number of viable cells, so the number of living cells can be determined by measuring ATP content. This kit employs an ATP-dependent, luciferase-catalyzed luminescence reaction to determine intracellular ATP content via a chemiluminescent signal, thereby assessing cell viability or quantifying viable cell numbers with high sensitivity and a wide linear range. Thanks to the high-purity luciferin substrate, a thermostable luciferase and optimized reaction reagents, this product provides stronger cell lysis capability and is suitable for cell viability analysis of 3D microtissue cultures. Adding the reagent to the cell culture lyses cell clusters and releases ATP, generating a stable luminescent signal. Within a certain range, luminescence intensity is proportional to the amount of ATP, i.e., the number of viable cells, enabling quantitative detection of viable cells.
Intended Use Cell viability analysis and quantification of viable cells in 3D microtissue cultures, spheroids and organoids by measuring intracellular ATP content.
Principle / Technology ATP-dependent luciferase-catalyzed luminescence: the reagent lyses 3D cell clusters and releases ATP, and the resulting luminescent signal is proportional, within a certain range, to the amount of ATP and thus to the number of viable cells. The formulation contains a high-purity luciferin substrate, a thermostable luciferase and optimized reaction reagents for enhanced lysis of 3D cultures.
Detection Method Luminescence (luminescence readout in kinetic mode on a luminescence-capable plate reader).
Sample Type 3D cell cultures, including spheroids, 3D microtissues and organoids (e.g., HeLa spheroids; ovarian cancer, lung cancer and thyroid cancer organoid models).
Performance Range / Specifications Convenient and fast: the detection reagent supplied in the kit is ready-to-use, with stable readings and rapid detection.
High sensitivity: provides a higher signal-to-noise ratio for 3D microtissue detection.
Efficient workflow: data can be recorded within 30 min or less after reagent addition.
Strong lysis capability: the optimized reaction reagents efficiently lyse cell clusters, making the kit suitable for cell viability analysis of 3D microtissue cultures.
High throughput: compatible with both low-sample-number detection and high-throughput screening of large sample numbers.
Reaction Conditions / Protocol Representative protocol for 3D microspheroids: culture HeLa cells at 5,000 cells per well in 100 uL in a low-attachment 96-well plate for 4-6 days; add 100 uL of detection reagent, shake for 5 min, let stand for 25 min, then transfer 100 uL to a white opaque-bottom 96-well plate and record the luminescence signal (kinetic readout).
Components / Formulation Ready-to-use 3D ATP detection reagent (high-purity luciferin substrate, thermostable luciferase and optimized lysis reagents; the detailed formulation is proprietary).
Storage Conditions Store at -25 to -15 °C, protected from light.
Shelf Life 1 year

Product Form Liquid, ready-to-use detection reagent.
Expiration Date / Stability Valid for 1 year when stored at -25 to -15 °C protected from light. Tested stability shows approximately 10% activity loss after 60 days at 4 °C, and 10 repeated freeze-thaw cycles have almost no effect on product activity.
Regulatory / Compliance For research use only.
Compatibility Compatible with both low-sample-number detection and high-throughput screening; suitable for 96-well low-attachment culture plates and white opaque-bottom plates for luminescence reading.
Application Notes / Precautions Performance has been benchmarked against mainstream commercial products in microspheroid and organoid models (ovarian, lung and thyroid cancer organoid models).
Differences from the ATP Luminescent Cell Viability Assay Kit 2.0: 1) Lysis components differ - this kit is optimized for 3D cells with stronger lysis capability, and the detailed formulation is proprietary. 2) Applicable sample types differ - the ATP Luminescent Cell Viability Assay Kit 2.0 shows good linearity in the range of 100 to 20,000 cells (the upper detection limit varies among cell types), whereas this kit targets cell clusters and has no defined cell number range.

For research use only, not for clinical use.
