Product Name Magnetic Bead-Based Circulating RNA Extraction Kit
Catalog No. DREK-0028
Description Magnetic bead-based kit for extraction of circulating cell-free RNA (cfRNA) including mRNA, miRNA, and other small non-coding RNAs from human plasma, serum, and other biofluids. Optimized for low-input, fragmented nucleic acids with carrier RNA-assisted binding. Extracted cfRNA is suitable for liquid biopsy applications including circulating transcriptome analysis, miRNA biomarker quantification, and non-invasive prenatal testing (NIPT).
Intended Use Circulating cfRNA extraction from plasma/serum; liquid biopsy transcriptome and miRNA analysis; cancer biomarker discovery; non-invasive prenatal testing; cardiovascular disease biomarker studies; exosome RNA extraction.
Principle / Technology Proteinase K digestion releases protein-bound RNA; optimized binding buffer with carrier RNA enhances recovery of low-concentration cfRNA; magnetic silica beads capture RNA; ethanol washes remove contaminants; elution in small volume maximizes concentration for downstream analysis
Detection Method Digest 0.5-4 mL plasma with proteinase K (60 °C, 20 min); add binding buffer with carrier RNA and magnetic beads; incubate 10 min with mixing; magnetic separate; wash 2×; dry beads briefly; elute in 20-50 µL
Sample Type Human plasma (EDTA preferred, citrate acceptable), serum, urine, CSF, bronchoalveolar lavage, ascites fluid; 0.5-4 mL input volume
Sensitivity / LOD Recovers circulating RNA from 0.5 mL plasma; detects miRNA at <100 copies/mL plasma; linear recovery across 0.5-4 mL input range; carrier RNA enhances recovery 3-5 fold for low inputs
Specificity Total RNA capture (mRNA, miRNA, lncRNA, piRNA); DNA carryover <1% (DNase I treatment optional); poly(A) carrier RNA does not interfere with downstream miRNA detection
Reaction Conditions / Protocol Proteinase K digestion: 20 min 60 °C; binding: 10 min; washes: 2× 5 min; elution: 5 min; total ~50 min for up to 24 samples
Components / Formulation Proteinase K (20 mg/mL), cfRNA binding buffer concentrate with carrier poly(A) RNA, silica magnetic beads (50 mg/mL), wash buffer 1, wash buffer 2, nuclease-free elution water, LoBind collection tubes (1.5 mL)
Storage Conditions Proteinase K at -20 °C; remaining components at 15-25 °C
Shelf Life 12 months; proteinase K 12 months at -20 °C
