Description A second-generation lentivirus packaging system consisting of three plasmids (transfer vector with transgene, packaging plasmid psPAX2, envelope plasmid pMD2.G) for the production of replication-incompetent lentiviral particles in HEK293T cells.
Intended Use Production of high-titer, VSV-G pseudotyped lentiviral particles for stable gene delivery into dividing and non-dividing mammalian cells, suitable for gene overexpression, shRNA knockdown, and CRISPR sgRNA library delivery.
Principle / Technology The viral packaging genes (gag, pol, rev, tat) are separated onto the packaging plasmid, while VSV-G envelope is provided on a separate plasmid, increasing biosafety; the transfer plasmid contains only the transgene flanked by LTR sequences, ensuring replication incompetence of packaged virions.
Detection Method Lentiviral titer by p24 ELISA, functional titer by antibiotic resistance colony formation or GFP expression in target cells
