| Reaction Conditions / Protocol | Use one dried dose consisting of two beads with 5 µL 10× primer mix and DNA template to a total volume of 50 µL. The 10× primer mix contains 16 µM FIP/BIP, 2 µM F3/B3 and 4 µM Loop F/B. Incubate at 65°C for 30-45 minutes and extend the time if needed. Read yellow as positive and red as negative. |

| Application Notes / Precautions | Optimize the reaction from 62°C to 68°C if required and avoid prolonged air exposure. Because the red-to-yellow readout depends on pH, do not use Tris-containing nucleic-acid storage solution; water is recommended. Prepare reactions in a clean bench and add template in a separate room or ventilated enclosure. |
