| Principle / Technology | A UDG/dUTP system degrades uracil-containing carryover products before amplification, reducing contamination risk. |
| Reaction Conditions / Protocol | For a 50 µL reaction, combine 12.5 µL 4× qPCR buffer, 1 µL 50× enzyme mix, each primer at 0.1-1.0 µM, hydrolysis probe at 0.05-0.25 µM, DNA template and water to volume. Standard program: 37°C for 2 minutes, 95°C for 1 minute, then 45 cycles of 95°C for 10 seconds and 60°C for 30 seconds. Fast program: 37°C for 2 minutes, 95°C for 30 seconds, then 42 cycles of 95°C for 5 seconds and 60°C for 10 seconds. |
| Components / Formulation | 4× qPCR buffer and 50× enzyme mix. The 100-, 1,000- and 10,000-reaction formats contain 1.25 mL, 12.5 mL and 125 mL buffer, plus 100 µL, 1 mL and 10 mL enzyme mix, respectively. |
