Product Name Dendrimer-Based Transfection Reagent, Activated Polyamidoamine
Catalog No. ATR-CTR-0016
Description An activated polyamidoamine dendrimer transfection reagent with a highly branched spherical architecture providing multiple DNA-binding sites for efficient gene delivery in a defined, non-lipid chemical delivery system.
Intended Use Transient and stable transfection of plasmid DNA into a broad range of mammalian cell types; particularly useful for cells sensitive to lipid-based reagents or where lipid-induced gene expression artifacts must be avoided.
Principle / Technology Activated PAMAM dendrimers (generation 5–7) possess a high density of terminal amine groups for DNA binding and a defined molecular architecture; the spherical shape provides a consistent compact structure for reproducible transfection; the proton-sponge effect of internal tertiary amines facilitates endosomal escape.
Detection Method Reporter gene expression by luciferase assay, cell viability by MTT or metabolic assay, gene expression profiling to detect lipid-induced artifacts
Sample Type Broad range of mammalian cell lines, primary cells, cells sensitive to lipid-based transfection reagents
Performance Range / Specifications Transfection efficiency 30–70% for standard cell lines; 20–50% for primary cells; cell viability >80% at optimal ratio
Sensitivity / LOD N/A
Specificity N/A
Reaction Conditions / Protocol Dilute DNA in serum-free medium; activate dendrimer per protocol; mix activated dendrimer with DNA; incubate 15 min; add to cells; replace medium after 4–24 hours; assay 24–72 hours.
Components / Formulation Activated PAMAM dendrimer solution in methanol, sterile water for reconstitution, detailed activation protocol
Storage Conditions 2–8°C, protect from light and moisture
Shelf Life 12 months
Package Specifications 100 transfections (24-well), 500 transfections
Product Form Liquid concentrate requiring activation
Quality Control Dendrimer generation and purity by MALDI-TOF, transfection efficiency validation with reference cell panel, cytotoxicity, endotoxin
Key Features Defined chemical structure; non-lipid chemistry; consistent particle size; minimal gene expression artifacts; broad cell compatibility; activated form for immediate use
