| Description | Ready-to-use DEAE-dextran (diethylaminoethyl-dextran) transfection reagent solution for efficient transient nucleic acid delivery into COS-7, CV-1, BSC-1, and other African green monkey kidney-derived cell lines. DEAE-dextran is a cationic polymer that complexes with negatively charged DNA to form positively charged particles that bind to the cell surface via electrostatic interactions with anionic proteoglycans. Uptake occurs through endocytosis, enhanced by a brief DMSO or glycerol shock step that permeabilizes endosomal membranes to release DNA into the cytoplasm. This method is particularly efficient for COS cell lines expressing SV40 large T-antigen, enabling episomal replication of SV40 origin-containing plasmids for very high expression levels. |

For chloroquine treatment: add 100 µM chloroquine during transfection incubation to inhibit lysosomal degradation. SV40 ori-containing plasmids amplify to very high copy number in COS-7 cells — 1-10 µg protein per 10^6 cells achievable. DEAE-dextran is not suitable for stable transfection. |
