Description Caspase (Cysteine-requiring Aspartate Protease) is a family of proteases that plays a crucial role in the apoptosis process. Caspase 1, also known as interleukin-1b converting enzyme (ICE), is sometimes written as caspase-1 or caspase 1. It is the only caspase in the caspase family capable of cleaving the IL-1b precursor protein or IL-18 precursor to produce the corresponding mature cytokines. Caspase 11 can cleave the 45 kDa caspase 1 precursor protein, producing 20 kDa and 10 kDa fragments. These two fragments can form heterodimers, which further assemble into tetramers. Caspase 1 can regulate apoptosis by cleaving the apoptotic Bcl-XL and modulate related immune responses by cleaving certain cytokine precursors.
Application This product uses spectrophotometry to detect caspase 1 enzyme activity in cell or tissue lysates or purified caspase 1 enzyme activity.

Notes Ac-YVAD-pNA should be avoided from repeated freeze-thaw cycles. Please note to aliquot appropriately. It is recommended to dilute the sample with water at a 1:1 ratio before determining protein concentration using the Bradford method to minimize interference from DTT on protein concentration measurement. pNA (Chinese name: 4-nitroaniline) is toxic to humans. Handle with extreme caution and ensure proper protective measures to avoid direct contact with the skin or inhalation. pNA (10 mM) may solidify and adhere to the bottom, walls, or lid of centrifuge tubes at low temperatures such as 4°C or ice bath conditions. It can be warmed in a water bath at 20–25°C for a short time until completely dissolved before use. For your safety and health, please wear a lab coat and disposable gloves during handling.
