Description A heat-based acid-fast staining kit using carbol fuchsin, acid alcohol decolorizer, and methylene blue counterstain for the microscopic detection of Mycobacterium species and other acid-fast organisms.
Intended Use Detection of Mycobacterium tuberculosis and other acid-fast bacilli in clinical sputum smears, tissue sections, and culture isolates for tuberculosis diagnosis and non-tuberculous mycobacteria identification.
Principle / Technology Carbol fuchsin with phenol penetrates the mycolic acid-rich waxy cell wall of mycobacteria upon heating; acid alcohol fails to decolorize mycobacteria due to their lipid-rich wall; methylene blue counterstains non-acid-fast background cells.
Detection Method Bright-field light microscopy at 100x oil immersion; fluorescence option available with auramine-rhodamine variant
