| Principle / Technology | Carbol fuchsin with phenol penetrates the mycolic acid-rich waxy cell wall of mycobacteria upon heating; acid alcohol fails to decolorize mycobacteria due to their lipid-rich wall; methylene blue counterstains non-acid-fast background cells. |
