Description An enhanced chemiluminescence (ECL) substrate kit for the sensitive detection of horseradish peroxidase (HRP)-conjugated antibodies in Western blotting applications. The two-component system produces an intense, long-lasting luminescent signal.
Intended Use Detection of HRP-labeled secondary antibodies in Western blot membrane-based immunodetection of target proteins extracted from cells or tissues.
Principle / Technology HRP catalyzes the oxidation of luminol in the presence of hydrogen peroxide to produce an excited-state intermediate that emits light at 425 nm upon decay to the ground state.
Detection Method Chemiluminescence detection using X-ray film, CCD camera, or digital imaging system
Sample Type PVDF or nitrocellulose membrane with immobilized proteins and HRP-conjugated antibodies
Performance Range / Specifications Detection sensitivity: low-picogram to high-femtogram range of target protein, depending on antibody affinity
Sensitivity / LOD ≤1 pg of target protein (with optimized primary and secondary antibodies)
Specificity Signal generation is dependent on the presence of HRP enzyme; negligible non-specific background when proper blocking and washing steps are performed
Reaction Conditions / Protocol Mix equal volumes of luminol/enhancer solution and peroxide solution, apply to membrane, incubate for 1–5 minutes, drain excess reagent, expose to film or imaging system
