| Description | The presence of protein in urine is called proteinuria, or urinary protein. Under normal conditions, urine contains only trace amounts of protein, and the urine protein test result should be negative. When kidney disease or injury occurs, there is often a persistent increase in protein levels in urine, resulting in a positive urine protein test result. Changes in protein levels are commonly used as an important basis for diagnosing kidney disease and assessing the extent of damage. |
| Application | Adding a protein precipitant and Alizarin Red dye to urine samples forms a protein-dye complex, which further dissolves in an alkaline solution. The change in absorbance at 560 nm can be used to quantitatively detect the content of urinary protein. |

| Notes | During centrifugation to collect the precipitate, all supernatant should be completely removed, and care should be taken not to aspirate the precipitate, which could cause loss and affect the test results. After adding Reagent 2, thoroughly vortex to ensure complete dissolution of the precipitate before measuring the absorbance value. If the ∆A measurement is greater than 0.3, it is recommended to dilute the sample with distilled water before re-measuring. If the ∆A measurement is less than 0.01 or the A measurement is close to the A blank, it is recommended to increase the sample volume before re-measuring and adjust the calculation accordingly. |
