Description The presence of protein in urine is referred to as proteinuria, or urinary protein. Under normal circumstances, urine contains only trace amounts of protein, and the qualitative result of a urine protein test should be negative. However, in cases of kidney disease or injury, there is often a persistent increase in the amount of protein in the urine, and the qualitative result of a urine protein test will be positive. Changes in protein levels are often used as an important basis for diagnosing kidney disease and determining the extent of damage.
Application Adding a protein precipitant and Lithmus Red dye to urine samples forms a protein-dye complex, which can be further dissolved in an alkaline solution. The change in absorbance at 560 nm can be used to quantitatively detect the content of urine protein.

Notes During centrifugation to collect the precipitate, all supernatant should be completely removed, and care should be taken not to aspirate the precipitate, which could cause loss and affect the experimental results. After adding reagent two, thoroughly shake to completely dissolve the precipitate before measuring the absorbance value; If the ∆A measurement is greater than 0.3, it is recommended to dilute the sample with distilled water before measuring; If the ∆A measurement is less than 0.01 or the A measurement is close to the A blank, it is recommended to increase the sample volume appropriately before measuring, and adjust the calculation accordingly.
