| Description | The sperm morphology staining kit (Shorr method) operates on a principle similar to that of the Papanicolaou staining method. Since proteins with different isoelectric points within sperm and cells carry different charges at the same pH, they selectively bind to corresponding dyes and become stained. The nucleus is composed of acidic substances and has a strong affinity for basic dyes; conversely, the cytoplasm contains basic substances and has a greater affinity for acidic dyes. It is particularly suitable for staining sperm and can also be used for staining cellular samples such as pleural fluid, ascites, and sputum. |
| Test Procedure | Fix the cell smears in a 95% ethanol–glacial acetic acid (3:1) fixative for 10–15 minutes. Soak in 80%, 70%, and 50% ethanol solutions for 1 minute each. Soak or rinse in distilled water or tap water for 1 minute. Stain with hematoxylin solution for 3–5 minutes. Rinse with tap water for 2 minutes. (Optional) Decolorize with 0.5% hydrochloric acid-ethanol decolorizing solution for 4–5 seconds. Rinse with tap water for 2 minutes. Blue with the blueing solution for 4 minutes. Rinse with tap water for 2 minutes. Add scarlet orange yellow staining solution dropwise and stain for 1–2 minutes. Rinse briefly with distilled water, then add the mordant solution dropwise and treat for 1–2 minutes. Add solid green staining solution and stain for 4 minutes. After decanting excess staining solution, immediately add 1% glacial acetic acid and decolorize for 30 seconds. Wash with water, then dehydrate with 50%, 70%, 80%, 95%, and anhydrous ethanol. Clear with xylene and mount with neutral gum. |
