Description Custom synthesis of allele-specific PCR primers for SNP genotyping by AS-PCR (Allele-Specific PCR) or ARMS-PCR (Amplification Refractory Mutation System). Two forward primers are designed with the 3′-terminal nucleotide matching either the wild-type or variant allele, paired with a common reverse primer. Supplied desalted with optional HPLC purification for enhanced allele discrimination.

Principle / Technology Allele-specific forward primers have the discriminating nucleotide at the 3′-terminus; Taq DNA polymerase extends only when the 3′ base pairs correctly with the template; a mismatch at the 3′ end prevents amplification, enabling allele discrimination on agarose gel or qPCR
Detection Method Customer provides SNP sequence context with variant nucleotide position indicated; primers designed for optimal Tm 58-62 °C with 3′ mismatch discrimination; supplied as two forward allele-specific primers plus common reverse primer; desalted standard, HPLC optional
