| Product Name | Custom DNA Oligonucleotide Synthesis Service (100 nmol, PAGE Purified) |
| Catalog No. | PRIPRO-0003 |
| Description | A high-scale custom DNA oligonucleotide synthesis service delivering 100 nanomole yield with polyacrylamide gel electrophoresis (PAGE) purification. PAGE purification achieves the highest resolution separation of full-length oligonucleotides from truncated synthesis by-products, including single-base (n–1) deletions. This purification method is the gold standard for long oligonucleotides (>40 bases) and applications demanding single-nucleotide resolution. Each oligo is gel-eluted, desalted, and lyophilized. Quality is verified by analytical PAGE and mass spectrometry. Recommended for demanding applications including gene synthesis, site-directed mutagenesis with long primers, and electrophoretic mobility shift assays (EMSA). |
| Intended Use | Optimized for gene synthesis assembly, long-primer site-directed mutagenesis, EMSA probe preparation, antisense oligonucleotide research, and any application where n–1 contamination must be minimized. |
| Principle / Technology | Solid-phase phosphoramidite synthesis followed by denaturing PAGE separation based on length; full-length band excised and eluted |
| Detection Method | Analytical PAGE and MALDI-TOF mass spectrometry |
| Sample Type | Template DNA, plasmid DNA, or genomic DNA for downstream applications |
| Performance Range / Specifications | 5–120 bases; optimal purification for 30–100 nt range |
| Sensitivity / LOD | N/A — custom synthesis reagent |
| Specificity | PAGE purification removes n–1 and shorter species; ≥97% full-length product by analytical PAGE; single-base resolution achieved |
| Reaction Conditions / Protocol | Resuspend in nuclease-free water or TE buffer. Prepare 100 µM stock. For gene synthesis, use equimolar amounts of overlapping oligos at 1–10 µM each in assembly reaction. |
