Product Name siRNA Duplex Synthesis Service, Custom Sequence, HPLC-Purified
Catalog No. PRIPRO-0033
Description Custom synthesis of siRNA duplexes (21-23 nt) with symmetric or asymmetric 3′ overhangs (dTdT or UU) for RNA interference (RNAi) gene knockdown studies. Each siRNA duplex is HPLC-purified to >90% purity, annealed, and supplied as a ready-to-transfect solution or lyophilized. Includes a negative control (scrambled sequence) siRNA for experimental controls.
Intended Use Specific gene knockdown in mammalian cells; RNAi pathway research; functional genomics screening; target validation; siRNA therapeutic development; in vivo gene silencing studies.
Principle / Technology Chemically synthesized 21-23 nt RNA oligonucleotides with complementary sense and antisense strands; annealed to form siRNA duplex with 3′ dTdT overhangs recognized by Dicer and RISC; antisense strand guides RISC to complementary mRNA for cleavage and degradation
Detection Method Customer provides target gene mRNA sequence (RefSeq accession or FASTA); optimal siRNA sequences designed using proprietary algorithm considering specificity, thermodynamic asymmetry, and off-target potential; 3-4 siRNA sequences recommended per target for initial screening
Sample Type Customer provides gene name, RefSeq mRNA accession, or target mRNA sequence (≥100 bp); specify species (human, mouse, rat, etc.)
Components / Formulation siRNA duplex (sense + antisense strands, annealed), negative control siRNA (scrambled, non-targeting), siRNA resuspension buffer (nuclease-free, pH 7.4); 5-20 nmol per duplex standard scale
Storage Conditions Lyophilized: 4 °C or -20 °C; resuspended: -20 °C in aliquots; avoid repeated freeze-thaw; protect from RNase contamination
Shelf Life Lyophilized: 24 months at -20 °C; resuspended: 6 months at -20 °C (aliquoted), 2 weeks at 4 °C
Package Specifications 5 nmol, 10 nmol, 20 nmol, 50 nmol, 100 nmol per duplex; negative control included
Product Form Lyophilized siRNA duplex (annealed) or ready-to-use solution (100 µM in resuspension buffer)
Key Features HPLC-purified >90% full-length duplex; 3′ dTdT overhangs for enhanced stability; proprietary design algorithm minimizes off-targets; negative control siRNA included; endotoxin-tested (<0.1 EU/nmol) for in vivo applications; chemically modified options: 2′-O-methyl, 2′-F, phosphorothioate; typical delivery 5-7 business days
Purity HPLC-purified >90% duplex; individual strands >95% by HPLC; endotoxin <0.1 EU/nmol; duplex confirmed by non-denaturing PAGE
Concentration As specified in synthesis report; typically lyophilized or at 100 µM in TE
Activity / Unit Definition Primer efficiency 90–110% in qPCR; Tm within ±2 °C of calculated value
Molecular Weight As specified per MALDI-TOF certificate; within 0.1% of theoretical mass
Source / Origin Solid-phase phosphoramidite chemical synthesis
pH Range / Optimal pH Stable in TE buffer pH 7.0–8.0; avoid acidic conditions causing depurination
Shipping Conditions Ambient; lyophilized siRNA stable at RT 2 weeks; resuspended siRNA ships on cold pack; protect from RNase
Expiration Date / Stability 24 months lyophilized at -20 °C; 6 months resuspended at -20 °C aliquoted; use RNase-free technique for resuspension; discard if non-denaturing PAGE shows single-strand dissociation
Regulatory / Compliance For laboratory and research use only; RUO; siRNAs are synthetic — no biohazard; for therapeutic use, GMP-grade synthesis available upon request; ISO 9001
Compatibility Compatible with standard PCR, qPCR, and sequencing platforms
Recommended Buffer System Resuspend in nuclease-free water or TE buffer pH 8.0
Application Notes / Precautions Protect fluorescent probes from light; avoid repeated freeze-thaw cycles; aliquot for single use
Batch-to-Batch Consistency MALDI-TOF mass confirmation per lot; qPCR performance within 0.5 Ct of reference lot

For research use only, not for clinical use.
