Product Name SDS-PAGE Gel Preparation Kit (Tris-Glycine System)
Catalog No. PTR-HMM-0060
Description A complete reagent kit for the preparation of polyacrylamide gels for sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The kit includes all required buffers and reagents for resolving and stacking gel preparation using the standard Laemmli system.
Intended Use Preparation of denaturing polyacrylamide gels for protein separation by molecular weight in SDS-PAGE, a fundamental technique in protein biochemistry and molecular biology.
Principle / Technology Proteins are denatured by SDS and migrate through the polyacrylamide gel matrix under an electric field at rates inversely proportional to the logarithm of their molecular weight.
Detection Method Electrophoretic separation; subsequent visualization by Coomassie staining, silver staining, or Western blot transfer
Sample Type Denatured protein samples (1–100 μg per lane) in loading buffer containing SDS and reducing agent
Performance Range / Specifications Resolution range: 10–250 kDa (8% gel), 6–80 kDa (12% gel), 2–40 kDa (15% gel); adjustable acrylamide percentage for optimal resolution
Sensitivity / LOD Detection limit: ~10 ng per band (Coomassie stain), ~0.1 ng per band (silver stain) — dependent on staining method
Specificity Separation based on molecular weight; does not distinguish between protein isoforms with similar molecular weights
Reaction Conditions / Protocol Prepare resolving gel solution, pour between glass plates, overlay with water; after polymerization, prepare stacking gel solution, pour, insert comb; gels ready in approximately 1 hour
Components / Formulation 30% Acrylamide/Bis-acrylamide solution (29:1 ratio), 1.5 M Tris-HCl pH 8.8 (resolving gel buffer), 0.5 M Tris-HCl pH 6.8 (stacking gel buffer), 10% SDS solution, 10% ammonium persulfate (APS), TEMED, detailed protocol
Storage Conditions 2–8°C; APS solution should be prepared fresh weekly; TEMED stored at room temperature in dark; acrylamide solution is light-sensitive
Shelf Life 12 months (acrylamide solution); 24 months (buffer components)
Package Specifications Sufficient for 40–60 mini-gels (8 x 10 cm)
Product Form Liquid reagents with APS as crystalline powder (reconstitute before use)
Quality Control Acrylamide solution tested for polymerization efficiency; each lot validated by SDS-PAGE with molecular weight markers; endotoxin tested for cell culture-related applications
Key Features Adjustable gel percentage for customized protein resolution; pre-mixed acrylamide/bis-acrylamide eliminates weighing steps; all buffers pH-adjusted and ready to use

For research use only, not for clinical use.
