Product Name Resazurin Cell Viability and Cytotoxicity Assay Kit
Catalog No. CATR-HMM-0045
Description A fluorometric and colorimetric assay kit based on the reduction of resazurin to resorufin by metabolically active cells. This optimized formulation includes an extraction and stabilization buffer that enhances signal intensity and duration for endpoint viability quantification and cytotoxicity screening.
Intended Use Endpoint measurement of cell viability for high-throughput screening of compound libraries, toxicology assessment of environmental chemicals, and in vitro biocompatibility testing of medical device materials.
Principle / Technology Resazurin (oxidized, blue, non-fluorescent) is reduced by mitochondrial and cytoplasmic reductases to resorufin (reduced, pink, highly fluorescent at Ex/Em: 530/590 nm); the rate and extent of reduction correlates with the metabolic capacity of viable cells; kit formulation stabilizes the resorufin signal for batch processing.
Detection Method Fuorescence (Ex/Em: 530/590 nm) or absorbance at 570 nm (reference 600 nm); compatible with fluorescence microplate readers and spectrophotometers
Sample Type Mammalian cells (transformed and primary), yeast, bacteria, and biofilm cultures; organotypic tissue slices; 3D spheroids after homogenization
Performance Range / Specifications Linear: 0.1–100% viability relative to untreated control; stable endpoint signal maintained for up to 4 hours after stopping the reaction
Sensitivity / LOD Detection of approximately 2% change in viability relative to untreated controls
Specificity Resazurin is reduced by diverse reductase enzyme systems; no signal from acellular medium; kit formulation minimizes precipitation and adsorption artifacts; compatible with PMMA and polystyrene labware
Reaction Conditions / Protocol Equilibrate all reagents to room temperature; combine Resazurin Stock with Assay Buffer (1:10 ratio); add 20 μL working solution per 100 μL medium; incubate 1–4 hours at 37°C; add Stop Solution; measure fluorescence at Ex/Em 530/590 nm
Components / Formulation Resazurin Stock Solution (100× concentrate), Assay Buffer (PBS-based with stabilizers), Stop Solution (SDS-based extraction buffer), Resorufin reference standard for calibration curve
Storage Conditions 2–8°C protected from light; Resazurin Stock stable for 12 months; reconstituted working solution should be used within 24 hours
Shelf Life 12 months from date of manufacture
Package Specifications 500 tests, 2,000 tests (96-well format)
Product Form Concentrated stock solution with separate buffer and stop solution components
Quality Control Each lot tested with positive (HeLa) and negative (heat-killed) cell controls; resorufin fluorescence standard curve provided; linearity across 0–100% viability range validated
Key Features Dual-mode detection (fluorometric and colorimetric) for compatibility with diverse plate reader equipment; stabilized endpoint signal for batch processing; resorufin reference standard included for instrument calibration
