Description Histochemical staining kit based on Perls' Prussian blue reaction for detection of ferric iron (Fe³⁺) in tissue sections, bone marrow aspirates, and cytology preparations. Ferric iron reacts with potassium ferrocyanide in acidic conditions to form insoluble Prussian blue pigment (ferric ferrocyanide), visible as bright blue deposits at sites of iron accumulation. Essential for hemochromatosis diagnosis and iron metabolism studies.
Intended Use Histological detection of ferric iron in tissue sections; bone marrow iron store assessment (sideroblastic anemia); hemochromatosis and hemosiderosis diagnosis; iron oxide nanoparticle biodistribution studies; liver biopsy iron grading.
Principle / Technology Perls' reaction: Fe³⁺ ions released by HCl react with potassium ferrocyanide to form insoluble ferric ferrocyanide (Prussian blue, Fe₄[Fe(CN)₆]₃); nuclear fast red counterstain provides contrast
Detection Method Deparaffinize and hydrate sections; apply potassium ferrocyanide/HCl mixture 20-30 min; wash in distilled water; counterstain with nuclear fast red 5 min; dehydrate, clear, and mount
