Product Name Pierce BCA Protein Assay Kit (Reducing Agent Compatible)
Catalog No. PTR-HMM-0075
Description A bicinchoninic acid protein assay kit specifically formulated for compatibility with samples containing reducing agents and detergents. The assay provides accurate protein quantification even in the presence of DTT, TCEP, and β-mercaptoethanol.
Intended Use Protein concentration determination in samples that contain reducing agents (up to 5 mM DTT, 10 mM TCEP, or 35 mM β-ME) and detergents, which interfere with other protein assay methods.
Principle / Technology Proteins reduce Cu2+ to Cu+ in an alkaline medium; the resulting Cu+ ions chelate with bicinchoninic acid to form a purple-colored complex with strong absorbance at 562 nm. The proprietary compatibility reagent neutralizes the effect of reducing agents.
Detection Method Colorimetric measurement at 562 nm
Sample Type Protein samples containing reducing agents, detergents, and other additives; compatible with cell lysis buffers containing up to 5 mM DTT or 2% SDS
Performance Range / Specifications Linear working range: 125–2000 μg/mL; sample volume: 25 μL (microplate) or 100 μL (cuvette)
Sensitivity / LOD ≥25 μg/mL lower detection limit
Specificity Universal protein detection with reduced protein-to-protein variation; the included compatibility reagent minimizes interference from reducing agents and common buffer components
Reaction Conditions / Protocol Add compatibility reagent to sample and incubate for 15 minutes at room temperature, add BCA working reagent, incubate at 37°C for 30 minutes (or room temperature for 2 hours), cool, and measure absorbance at 562 nm
Components / Formulation BCA Reagent A, BCA Reagent B, compatibility reagent, BSA standard set (2 mg/mL), microplate protocol, cuvette protocol
Storage Conditions BCA reagents at 2–8°C; compatibility reagent at room temperature; working reagent must be prepared fresh
Shelf Life 24 months for unopened reagents
Package Specifications 500 tube assays or 5000 microplate assays
Product Form Liquid reagents
Quality Control Each lot validated with standard BSA curves in the presence and absence of reducing agents; linearity and accuracy verified; endotoxin tested
Key Features Unique compatibility reagent formulation enables accurate protein measurement in reducing agent-containing samples, eliminating the need for sample dialysis or precipitation prior to quantitation

For research use only, not for clinical use.
