| Principle / Technology | Periodic acid oxidizes vicinal glycol groups in polysaccharides to aldehydes; Schiff's reagent (fuchsin-sulfurous acid) reacts with aldehydes to produce a magenta-colored product; diastase digestion control differentiates glycogen from other PAS-positive substances. |
