Description A histochemical staining kit employing periodic acid oxidation followed by Schiff's reagent for the detection of glycogen, neutral mucopolysaccharides, basement membranes, and fungal organisms in tissue sections.
Intended Use Demonstration of glycogen in liver and muscle biopsies; detection of neutral mucins in gastrointestinal and respiratory tract specimens; identification of fungal organisms (Candida, Aspergillus) in tissue; visualization of basement membrane pathology in renal biopsies.
Principle / Technology Periodic acid oxidizes vicinal glycol groups in polysaccharides to aldehydes; Schiff's reagent (fuchsin-sulfurous acid) reacts with aldehydes to produce a magenta-colored product; diastase digestion control differentiates glycogen from other PAS-positive substances.
Detection Method Bright-field light microscopy
