Description A 10% aqueous nigrosin solution for negative staining of bacterial capsules and spores, providing a dark background against which unstained structures appear as clear halos or bright refractile bodies.
Intended Use Demonstration of bacterial capsules (Klebsiella pneumoniae, Streptococcus pneumoniae, Cryptococcus neoformans) and endospores (Bacillus, Clostridium species) by negative staining in microbiology teaching and clinical laboratories.
Principle / Technology Nigrosin, an acidic aniline dye, does not penetrate bacterial cells or capsules due to charge repulsion; it forms a dark background film against which the unstained capsule appears as a clear zone and spores as bright refractile oval bodies.
Detection Method Bright-field light microscopy at 100x oil immersion
