| Product Name | Neutral Red Uptake Cytotoxicity Assay Kit |
| Catalog No. | CATR-HMM-0046 |
| Description | A colorimetric assay based on the ability of viable cells to incorporate and bind the supravital dye Neutral Red in their lysosomes. This OECD-validated test method is widely applied in regulatory toxicology for assessing the basal cytotoxicity of chemicals and consumer product ingredients. |
| Intended Use | In vitro basal cytotoxicity testing of chemicals, cosmetics, pharmaceuticals, and medical devices according to OECD Test Guideline 432 for regulatory safety assessment. |
| Principle / Technology | Neutral Red (3-amino-7-dimethylamino-2-methylphenazine hydrochloride), a weak cationic dye, penetrates cell membranes by non-ionic passive diffusion and accumulates in lysosomes of viable cells where the acidic pH traps the protonated form; after washing and dye extraction, absorbed color is proportional to the number of viable cells. |
| Detection Method | Colorimetric absorbance measurement at 540 nm using a microplate spectrophotometer |
| Sample Type | BALB/c 3T3 mouse fibroblasts (OECD TG 432 standard), normal human keratinocytes, HepG2 hepatoma cells, and other adherent mammalian cell lines for regulatory cytotoxicity screening |
| Performance Range / Specifications | Linear detection: 5,000–50,000 cells per well for 3T3 cells; IC50 determination across 6–8 point concentration range; absorbance 0.1–1.5 OD at 540 nm |
| Sensitivity / LOD | Detection of approximately 10% reduction in cell viability relative to untreated controls for IC50 determination |
| Specificity | Neutral Red uptake is specific to lysosomal accumulation in living cells; fixed or dead cells cannot retain the dye through the washing steps; validated per OECD guidelines for regulatory use |
| Reaction Conditions / Protocol | Expose cultured cells to test substances for 24–48 hours; wash with PBS; add Neutral Red medium (25–50 μg/mL) and incubate 3 hours at 37°C; wash carefully to remove extracellular dye; add Neutral Red desorb solution (1% acetic acid in 50% ethanol); shake 10–45 minutes; read absorbance at 540 nm |
| Components / Formulation | Neutral Red stock solution (lyophilized or 4 mg/mL), Neutral Red Medium (diluted in complete culture medium), Neutral Red Desorb Solution (acidified ethanol), PBS washing solution, positive control (sodium lauryl sulfate standard) |
| Storage Conditions | Neutral Red stock: 2–8°C protected from light for 12 months; Neutral Red medium prepared fresh before each use; Desorb Solution: room temperature 24 months |
| Shelf Life | Neutral Red stock: 12 months; other kit components: 24 months |
| Package Specifications | 96-well format for 500–2,000 test substance evaluations |
| Product Form | Lyophilized dye with separate desorb and wash solutions |
| Quality Control | Each lot validated using OECD TG 432 reference chemicals (sodium lauryl sulfate) with established IC50 acceptance range; inter-plate CV <15%; Neutral Red purity ≥90% by HPLC |
| Key Features | OECD-validated and GLP-compliant method for regulatory submission; widely harmonized across international chemical safety programs; simple equipment requirements; cost-effective for routine toxicology laboratories |
For research use only, not for clinical use.
