Description A colorimetric assay based on the ability of viable cells to incorporate and bind the supravital dye Neutral Red in their lysosomes. This OECD-validated test method is widely applied in regulatory toxicology for assessing the basal cytotoxicity of chemicals and consumer product ingredients.
Intended Use In vitro basal cytotoxicity testing of chemicals, cosmetics, pharmaceuticals, and medical devices according to OECD Test Guideline 432 for regulatory safety assessment.
Principle / Technology Neutral Red (3-amino-7-dimethylamino-2-methylphenazine hydrochloride), a weak cationic dye, penetrates cell membranes by non-ionic passive diffusion and accumulates in lysosomes of viable cells where the acidic pH traps the protonated form; after washing and dye extraction, absorbed color is proportional to the number of viable cells.
Detection Method Colorimetric absorbance measurement at 540 nm using a microplate spectrophotometer
Sample Type BALB/c 3T3 mouse fibroblasts (OECD TG 432 standard), normal human keratinocytes, HepG2 hepatoma cells, and other adherent mammalian cell lines for regulatory cytotoxicity screening

Reaction Conditions / Protocol Expose cultured cells to test substances for 24–48 hours; wash with PBS; add Neutral Red medium (25–50 μg/mL) and incubate 3 hours at 37°C; wash carefully to remove extracellular dye; add Neutral Red desorb solution (1% acetic acid in 50% ethanol); shake 10–45 minutes; read absorbance at 540 nm
