Description A tetrazolium dye-based colorimetric kit designed to assess cellular metabolic activity as an indicator of cell viability, proliferation, and cytotoxicity. The yellow MTT reagent is reduced by mitochondrial dehydrogenases in metabolically active cells to form insoluble purple formazan crystals that can be solubilized and measured spectrophotometrically.
Intended Use Quantitative measurement of cell viability and proliferation in mammalian cells following drug treatment, growth factor stimulation, or cytotoxic exposure in research and drug discovery applications.
Principle / Technology Reduction of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) by NAD(P)H-dependent oxidoreductase enzymes in viable cells yields purple formazan crystals; dissolved crystals are measured by absorbance at 570 nm.
Detection Method Colorimetric absorbance measurement at 570 nm using a microplate reader, with background subtraction at 630 nm
