Description A homogeneous colorimetric method for assessing cell proliferation, viability, and chemosensitivity using the tetrazolium compound MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium). MTS is reduced by metabolically active cells in the presence of the electron coupling reagent phenazine methosulfate to generate a soluble formazan product measured at 490 nm.
Intended Use Non-radioactive quantification of metabolically active viable cells in growth factor bioassays, drug sensitivity testing, and large-scale cell-based screening campaigns.
Principle / Technology NAD(P)H-dependent cellular oxidoreductases reduce MTS tetrazolium to a soluble formazan product in the presence of the intermediate electron acceptor PMS (phenazine methosulfate); formazan absorbance at 490 nm is directly proportional to the number of living cells.
