| Product Name | MTS Cell Proliferation Colorimetric Assay Kit |
| Catalog No. | CATR-HMM-0041 |
| Description | A homogeneous colorimetric method for assessing cell proliferation, viability, and chemosensitivity using the tetrazolium compound MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium). MTS is reduced by metabolically active cells in the presence of the electron coupling reagent phenazine methosulfate to generate a soluble formazan product measured at 490 nm. |
| Intended Use | Non-radioactive quantification of metabolically active viable cells in growth factor bioassays, drug sensitivity testing, and large-scale cell-based screening campaigns. |
| Principle / Technology | NAD(P)H-dependent cellular oxidoreductases reduce MTS tetrazolium to a soluble formazan product in the presence of the intermediate electron acceptor PMS (phenazine methosulfate); formazan absorbance at 490 nm is directly proportional to the number of living cells. |
| Detection Method | Colorimetric absorbance measurement at 490 nm with a reference wavelength of 630 nm using a microplate spectrophotometer |
| Sample Type | Adherent and suspension mammalian cells, CTL assays, hybridoma cell screening; works with cells cultured in 96-well microplates |
| Performance Range / Specifications | Linear range: 500–80,000 cells per well depending on cell type; color development reaches plateau in 1–4 hours; signal stable for 18 hours at 37°C |
| Sensitivity / LOD | Approximately 500 viable cells per well for most mammalian cell lines |
| Specificity | MTS reduction activity reflects viable cell metabolic activity; dead cells produce negligible formazan; phenolic red in culture medium does not significantly interfere with 490 nm measurement against 630 nm reference |
| Reaction Conditions / Protocol | Thaw MTS and PMS solutions; mix 2 mL MTS with 100 μL PMS for each plate (prepare fresh); add 20 μL combined reagent per 100 μL culture medium; incubate 1–4 hours at 37°C with 5% CO2; read absorbance at 490 nm with 630 nm reference; no washing steps required |
| Components / Formulation | MTS Reagent (lyophilized or concentrated solution), PMS (phenazine methosulfate) solution, sterile PBS for preparation of working reagent |
| Storage Conditions | MTS reagent: -20°C protected from light; PMS solution: -20°C protected from light; MTS/PMS combined working reagent stable for 2 hours at room temperature in the dark |
| Shelf Life | 12 months from date of manufacture |
| Package Specifications | 1,000 assays, 5,000 assays (96-well format) |
| Product Form | Lyophilized MTS with separate PMS coupling reagent |
| Quality Control | Each lot validated against HeLa and NIH/3T3 cells; linearity (R2 ≥0.99) confirmed; endotoxin tested |
| Key Features | Homogeneous one-solution addition format without washing steps; water-soluble formazan eliminates organic solvent step; non-toxic reagent allows further cell analysis; suitable for automated high-throughput screening platforms |
