Product Name Micro BCA Protein Assay Kit, High-Sensitivity (0.5-20 µg/mL)
Catalog No. PTR-HMM-0088
Description Micro BCA (bicinchoninic acid) protein assay kit for ultrasensitive total protein quantification down to 0.5 µg/mL. The Micro BCA chemistry uses Cu⁺ reduction of BCA with tartrate as a stabilizing chelator at elevated temperature (60 °C), dramatically increasing sensitivity compared to standard BCA and enabling detection of dilute protein samples from immunoprecipitation eluates, column chromatography fractions, and low-cell-number samples.
Intended Use Ultrasensitive protein quantification (0.5-20 µg/mL); dilute protein sample quantification; immunoprecipitation eluate protein determination; chromatography fraction analysis; low-cell-number sample quantification; protein determination prior to mass spectrometry.
Principle / Technology Proteins reduce Cu²⁺ to Cu⁺ in alkaline medium (Biuret reaction); Cu⁺ chelates with 2 molecules of bicinchoninic acid (BCA) forming purple complex with strong absorbance at 562 nm; Micro BCA: tartrate stabilizes Cu⁺ at 60 °C enabling 10× sensitivity improvement over standard BCA
Detection Method Colorimetric; absorbance at 562 nm; incubation at 60 °C for 60 min; standard curve 0.5-20 µg/mL BSA
Sample Type Dilute protein solutions; aqueous buffers; samples with low protein concentration (<20 µg/mL); compatible with 0.5-5% detergents after dilution
Sensitivity / LOD Linear range 0.5-20 µg/mL BSA (standard protocol); detection limit 0.2 µg/mL; 10-fold more sensitive than standard BCA
Specificity Protein quantification based on peptide bond-mediated Cu²⁺ reduction and Cys/Tyr/Trp residue contribution; chelating agents (EDTA >10 mM), reducing agents (DTT >1 mM), and ammonium sulfate (>25% saturation) interfere
Reaction Conditions / Protocol Mix 0.5 mL sample with 0.5 mL Micro BCA working reagent (MA:MB:MC = 25:24:1); incubate at 60 °C for 60 min; cool to RT; read A562; compare to BSA standard curve
Components / Formulation Micro BCA Reagent MA (sodium carbonate, sodium tartrate, sodium bicarbonate in 0.1 M NaOH), Micro BCA Reagent MB (BCA disodium salt in water), Micro BCA Reagent MC (copper(II) sulfate pentahydrate 4% solution), BSA standard set (2.0, 1.5, 1.0, 0.75, 0.5, 0.25, 0.125, 0 mg/mL), instruction manual
Storage Conditions 15-25 °C; BSA standards at 4 °C; Reagent C (copper sulfate) stable at RT
Shelf Life 12 months from manufacture date
Package Specifications 500 assays, 1,000 assays; accommodates 1 mL cuvette or 200 µL microplate formats
Product Form Liquid reagents; BSA standard set pre-diluted
Key Features 10× more sensitive than standard BCA (0.5 vs. 5 µg/mL detection limit); compatible with common detergents (SDS, Triton X-100, NP-40) at working concentrations; simple mix-and-read protocol; pre-diluted BSA standards for convenience; 60 °C incubation minimizes time (1 hr vs. 2 hr for standard BCA at 37 °C)
Purity BCA reagent purity >99%; BSA standards fatty acid-free, >98% pure; copper sulfate ACS grade
Concentration As specified on product label or certificate of analysis
Activity / Unit Definition Binding capacity or enzymatic activity as specified per product documentation
Molecular Weight Varies by product as specified in documentation
Source / Origin Recombinant expression in E. coli, mammalian, or insect cell systems; synthetic chemistry for small molecule reagents
pH Range / Optimal pH pH 7.0–8.0 for most protein applications
Shipping Conditions Ambient; BSA standards cold pack
Expiration Date / Stability 12 months at RT for reagents; BSA standards 12 months at 4 °C; working reagent stable 24 hr at RT; discard Reagent MC if blue precipitate forms (Cu(OH)₂)
Regulatory / Compliance For laboratory and research use only; RUO; Reagent MA contains NaOH — corrosive; ISO 9001
Compatibility Compatible with standard SDS-PAGE, Western blot, mass spectrometry, and chromatography systems
Recommended Buffer System PBS, Tris, or HEPES buffers as specified in product protocol
Application Notes / Precautions Avoid repeated freeze-thaw cycles for protein solutions; use fresh aliquots; protect light-sensitive reagents
Batch-to-Batch Consistency Protein concentration and activity within ±15% of reference lot
