Description A polychrome methylene blue solution for bacteriological staining, demonstrating metachromatic granules in Corynebacterium diphtheriae (Albert's stain), and for reticulocyte enumeration in blood specimens.
Intended Use Microscopic detection of metachromatic granules (volutin) in Corynebacterium species; supravital staining of reticulocytes in peripheral blood; general bacterial morphology staining in microbiology.
Principle / Technology Methylene blue in alkaline borax solution forms polychrome oxidation products (azure dyes) that produce metachromatic staining of volutin granules (purple-red) against a blue bacterial cytoplasm.
Detection Method Bright-field light microscopy at 100x oil immersion
Sample Type Bacterial smears from culture plates, peripheral blood for reticulocyte count, throat swabs for Albert's stain
