Product Name Mammalian Protein Extraction Reagent (MPER)
Catalog No. PTR-HMM-0062
Description A gentle detergent-based protein extraction reagent formulated to efficiently lyse cultured mammalian cells while preserving the native conformation and activity of extracted proteins. This reagent avoids strong denaturants, making it ideal for functional studies.
Intended Use Extraction of native, functional proteins from cultured mammalian cells for applications requiring preserved protein activity, including enzyme assays, immunoprecipitation, and protein-protein interaction studies.
Principle / Technology Proprietary detergent formulation disrupts cell membranes without denaturing proteins, releasing soluble cytoplasmic and membrane-associated proteins while maintaining their native three-dimensional structure.
Detection Method Gentle lysis by incubation at room temperature or on ice, followed by centrifugation to recover soluble protein fraction
Sample Type Cultured adherent and suspension mammalian cells; optimized for common cell lines (HeLa, HEK293, CHO, Jurkat, 3T3)
Performance Range / Specifications Typical protein recovery: 2–5 mg/mL from 10^7 cells; extraction efficiency >90% for cytoplasmic proteins
Sensitivity / LOD Effective protein extraction from as few as 10^4 cells
Specificity Extracts cytoplasmic and membrane-associated proteins; nuclear proteins are partially extracted; suitable for kinase, phosphatase, and other enzymatic activity assays
Reaction Conditions / Protocol Wash cells with PBS, add extraction reagent (200 μL per 10^6 cells), incubate at room temperature for 5–10 minutes with gentle shaking, centrifuge at 14,000g for 10 minutes, collect supernatant containing soluble proteins
Components / Formulation Ready-to-use MPER reagent in Tris-buffered saline with proprietary detergent blend; optional phosphatase and protease inhibitor additives sold separately
Storage Conditions 2–8°C; protect from prolonged exposure to temperatures above 25°C
Shelf Life 18 months from date of manufacture
Package Specifications 250 mL (sufficient for approximately 1,250 extractions at 200 μL each)
Product Form Ready-to-use liquid reagent
Quality Control Each lot tested for lysis efficiency by protein recovery quantification and Western blot analysis of cytoplasmic (GAPDH), membrane (EGFR), and nuclear (Histone H3) protein markers; endotoxin <0.1 EU/mL
Key Features Native protein preservation for functional assays; rapid 5–10 minute protocol; no need for mechanical homogenization; low viscosity simplifies pipetting steps
