Description It is a reagent based on the INT colorimetric reaction catalyzed by lactate dehydrogenase, used to detect the activity of lactate dehydrogenase released during cytotoxicity testing via colorimetry or to detect lactate dehydrogenase activity in other samples. Cell membrane structural damage caused by apoptosis or necrosis leads to the release of enzymes from the cytoplasm into the culture medium, including lactate dehydrogenase (LDH), which has relatively stable enzymatic activity. By detecting the LDH activity released into the culture medium from cells with ruptured plasma membranes, quantitative analysis of cytotoxicity can be achieved. LDH release is considered an important indicator of cell membrane integrity and is widely used in cytotoxicity assays. LDH release is regarded as a safe and effective alternative to the previously used method of labeling cells with radioactive 51Cr and subsequently detecting cell membrane integrity via 51Cr release.

Notes Since serum contains lactate dehydrogenase, it is recommended that the concentration of serum used should not exceed 1%, and it is best to use heat-inactivated serum. Collect the LDH released in the cell supernatant, which can be placed in the refrigerator overnight and tested the next day. However, in order to reduce experimental errors, it is recommended that clients collect and test samples immediately. This product is generally used to calculate the mortality rate of the sample, not the mortality rate of the control. Additionally, the mortality rate of the sample should be calculated using the control.
