Product Name Immobilon-P PVDF Transfer Membrane (0.45 μm)
Catalog No. PTR-HMM-0080
Description A polyvinylidene difluoride membrane with 0.45 μm pore size optimized for Western blot protein transfer from SDS-PAGE gels. The membrane provides high protein binding capacity and mechanical strength for immunoblot detection workflows.
Intended Use Protein immobilization following SDS-PAGE for subsequent immunodetection by Western blotting using chemiluminescent, fluorescent, or chromogenic detection methods.
Principle / Technology Proteins are electrophoretically transferred from the polyacrylamide gel matrix onto the PVDF membrane, where they bind through hydrophobic and dipole interactions, creating a replica of the gel's protein separation pattern.
Detection Method Immunodetection with primary and secondary antibodies followed by ECL, fluorescent, or chromogenic visualization
Sample Type Proteins separated by SDS-PAGE or native PAGE
Performance Range / Specifications Protein binding capacity: 125–200 μg/cm² for IgG; pore size: 0.45 μm; applicable molecular weight range: >20 kDa; membrane dimensions: 10 cm × 15 cm sheets or 26.5 cm × 3.75 m roll
Sensitivity / LOD Detection sensitivity commensurate with the immunodetection system; typical detection limits in the low picogram range with optimized antibodies and ECL detection
Specificity Binds most proteins regardless of isoelectric point; hydrophobic PVDF surface provides strong protein retention during washing and reprobing cycles
Reaction Conditions / Protocol Pre-wet membrane in 100% methanol for 15 seconds, rinse in ultrapure water for 2 minutes, equilibrate in transfer buffer for at least 5 minutes, assemble transfer sandwich, transfer at 100V for 60 minutes (wet transfer) or according to semi-dry transfer protocol
Components / Formulation PVDF membrane sheets or rolls, hydrophobic surface requires methanol pre-wetting before aqueous buffer equilibration
Storage Conditions Room temperature; store in a dry environment; membrane sheets should remain in the protective interleaf paper until use
Shelf Life 36 months from date of manufacture
Package Specifications 10 sheets (10 cm × 15 cm) or 1 roll (26.5 cm × 3.75 m)
Product Form Flat PVDF membrane sheets or roll with interleaf paper
Quality Control Each lot tested for protein binding capacity using 125I-labeled IgG; wetting uniformity verified; background signal tested with standard ECL detection system
Key Features PVDF provides superior protein retention compared to nitrocellulose, enabling multiple stripping and reprobing cycles on a single membrane; the high mechanical strength allows convenient handling without tearing; 0.45 μm pore size provides optimal balance of binding capacity and flow characteristics
