Product Name GST-Tag Protein Purification Resin (Glutathione Agarose)
Catalog No. PTR-HMM-0070
Description A glutathione-coupled agarose resin for the affinity purification of recombinant proteins fused to glutathione S-transferase. The resin provides high-capacity capture of GST-tagged proteins from bacterial, yeast, and mammalian expression systems.
Intended Use Affinity purification of GST-fusion proteins from clarified cell lysates under native conditions for biochemical characterization, antibody production, and functional studies.
Principle / Technology Immobilized glutathione on cross-linked agarose beads binds specifically to the glutathione S-transferase affinity tag with high selectivity; bound GST-fusion proteins are eluted with reduced glutathione under mild, non-denaturing conditions.
Detection Method Column or batch-mode chromatography with UV detection at 280 nm
Sample Type Clarified lysates from cells expressing GST-fusion recombinant proteins; compatible with E. coli, yeast, and mammalian expression systems
Performance Range / Specifications Binding capacity: ≥10 mg of GST protein per milliliter of settled resin; typical elution purity exceeds 90% in a single step
Sensitivity / LOD Sufficient for purifying GST-fusion proteins from low-expression constructs with optimization of binding conditions
Specificity High selectivity for GST and GST-fusion proteins; minimal non-specific binding of host cell proteins under recommended buffer conditions
Reaction Conditions / Protocol Equilibrate column with PBS, load clarified cell lysate at a flow rate of 0.5–1 mL/min, wash column with 10–20 column volumes of PBS, elute with 50 mM Tris-HCl pH 8.0 containing 10 mM reduced glutathione, collect fractions and analyze by SDS-PAGE
Components / Formulation Glutathione Sepharose resin as a 50% slurry in 20% ethanol, included: 10× PBS concentrate, reduced glutathione elution buffer powder, empty chromatography columns
Storage Conditions 2–8°C; do not freeze the resin; storage solution contains 20% ethanol as preservative
Shelf Life 24 months
Package Specifications 10 mL of 50% slurry (5 mL bed volume)
Product Form 50% bead slurry in ethanol-containing storage buffer
Quality Control Binding capacity validated using purified recombinant GST protein; resin chemical stability tested; glutathione leaching measured by HPLC
Key Features Mild elution conditions preserve native protein conformation and enzymatic activity; high selectivity reduces purification steps; resin is compatible with standard laboratory chromatography equipment
