Product Name FluoroProfile Protein Quantification Kit (Fluorometric, 0.02-2 µg/mL)
Catalog No. PTR-HMM-0089
Description Ultrasensitive fluorometric protein quantification kit using a proprietary fluorogenic dye that fluoresces upon non-covalent binding to protein hydrophobic regions. With a detection range of 0.02-2 µg/mL, this assay is 250-fold more sensitive than standard BCA and 10-fold more sensitive than Micro BCA. Designed for quantifying extremely dilute protein samples from laser capture microdissection, single-cell proteomics preparations, and trace protein analysis.
Intended Use Ultrasensitive protein quantification of dilute samples; luminescence-based detection complement; laser capture microdissection (LCM) sample quantification; single-cell proteomics; trace protein determination in process impurities; protein quantification for ultrasensitive ELISA.
Principle / Technology Proprietary fluorogenic dye is essentially non-fluorescent in aqueous solution but fluoresces strongly (Ex/Em 485/530 nm) upon binding to hydrophobic regions of proteins; fluorescence intensity is proportional to protein concentration; minimal protein-to-protein variation due to broad hydrophobic region recognition
Detection Method Fluorometric; excitation 480-485 nm, emission 525-535 nm; 96-well or 384-well black plate format
Sample Type Aqueous protein solutions; cell/tissue lysates after high dilution; immunoprecipitation eluates; column fractions; samples with 0.02-2 µg/mL protein range
Sensitivity / LOD Detection 0.02-2 µg/mL BSA (250× more sensitive than BCA); linear dynamic range 2 orders of magnitude; 0.02 µg/mL is detectable above blank
Specificity Binds hydrophobic protein regions; less protein-to-protein variation than Coomassie dye-binding; free amino acids, peptides <3 kDa, and nucleic acids show minimal interference
Reaction Conditions / Protocol Add 10-100 µL dilute protein sample to black plate; add equal volume of FluoroProfile working reagent; mix gently; incubate 5-15 min at RT protected from light; read fluorescence Ex/Em 485/530 nm; compare to BSA standard curve
Components / Formulation FluoroProfile dye reagent (200× concentrate in DMSO), BSA protein standard (2 mg/mL), assay buffer (PBS pH 7.4), black 96-well plate (optional), adhesive plate seal
Storage Conditions 2-8 °C for dye reagent; -20 °C for long-term; protect from light; BSA standard at 4 °C
Shelf Life 12 months at -20 °C; 6 months at 4 °C; working reagent stable 2 hr at RT, 24 hr at 4 °C
Package Specifications 200 assays, 1,000 assays (96-well); 4,000 assays (384-well)
Product Form Liquid dye concentrate in DMSO; BSA standard in PBS
