Description Custom synthesis of locus-specific FISH probes for detection of chromosomal abnormalities, gene amplifications, deletions, and rearrangements by fluorescence microscopy. Probes are generated from BAC (Bacterial Artificial Chromosome) clones or long-range PCR products, fluorescently labeled with SpectrumGreen, SpectrumOrange, SpectrumRed, or SpectrumAqua fluorophores via nick translation. Each probe includes hybridization buffer for direct use.
Intended Use Detection of gene amplification (HER2, EGFR, MYCN, MET); chromosomal translocation detection (BCR-ABL, EML4-ALK, TMPRSS2-ERG); gene deletion detection (PTEN, CDKN2A, TP53); chromosomal enumeration for aneuploidy screening; break-apart and dual-fusion probe designs.
Principle / Technology BAC DNA or long-range PCR product is labeled with fluorescent-dUTP by nick translation; labeled probe DNA is ethanol-precipitated with Cot-1 DNA (blocks repetitive sequences) and resuspended in hybridization buffer; probe hybridizes to complementary chromosomal target on metaphase spreads or interphase nuclei; fluorescence microscopy visualizes signal at target locus
Detection Method Customer provides target genomic coordinates or gene name; probes designed from BAC clones spanning 100-300 kb at target locus; labeled with specified fluorophore; supplied as 200 µL hybridization-ready probe (20 tests at 10 µL/test)
