Product Name FLAG Peptide Competitive Elution Reagent
Catalog No. PTR-HMM-0071
Description A 3×FLAG peptide solution for the gentle competitive elution of FLAG-tagged recombinant proteins from anti-FLAG affinity resins. This elution method preserves protein activity by avoiding the harsh pH conditions used in conventional elution protocols.
Intended Use Gentle, non-denaturing elution of FLAG-tagged fusion proteins from anti-FLAG affinity purification resins, maintaining native protein conformation and biological activity.
Principle / Technology Synthetic 3×FLAG peptide (DYKDHDGDYKDHDIDYKDDDDK) competes with the FLAG epitope tag on the fusion protein for binding to the immobilized anti-FLAG monoclonal antibody, resulting in specific displacement and elution of the tagged protein.
Detection Method Elution under physiological pH and ionic strength conditions; eluted protein is recovered in a biologically active state
Sample Type FLAG-tagged recombinant proteins bound to anti-FLAG M2 or other FLAG-specific affinity resins
Performance Range / Specifications Effective elution achieved with 100–150 μg/mL 3×FLAG peptide in TBS buffer; typical elution volume is 2–5 column bed volumes
Sensitivity / LOD Complete elution of FLAG-tagged proteins at concentrations as low as 50 μg/mL
Specificity The 3×FLAG peptide sequence is specifically recognized by anti-FLAG monoclonal antibodies; it does not cross-react with other epitope tag systems (HA, Myc, V5, His)
Reaction Conditions / Protocol After binding FLAG-tagged protein to the anti-FLAG resin and washing, apply 3×FLAG peptide solution (150 μg/mL in TBS), incubate for 30 minutes at 4°C with gentle agitation, collect eluate, repeat if needed for complete recovery
Components / Formulation Lyophilized 3×FLAG peptide (sufficient for preparation of 50 mL of elution buffer), TBS buffer salts for elution buffer preparation
Storage Conditions Lyophilized peptide at -20°C desiccated; reconstituted peptide solution can be stored at 2–8°C for 2 weeks or aliquoted and frozen at -20°C for 3 months
Shelf Life 24 months for lyophilized peptide
Package Specifications 5 mg of lyophilized 3×FLAG peptide
Product Form White lyophilized powder
Quality Control Peptide purity >95% by HPLC; molecular weight confirmed by mass spectrometry; biological activity verified by competitive elution of FLAG-tagged protein standard from anti-FLAG resin
Key Features Gentle elution preserves protein activity and complex integrity for downstream functional assays; no residual peptide contamination when followed by dialysis or desalting; compatible with tandem affinity purification strategies
