Description An ultra-pure preparation of dithiothreitol for the reduction of disulfide bonds in proteins and peptides, used in SDS-PAGE sample preparation, protein refolding, and enzymatic reaction buffers.
Intended Use Reduction of disulfide bonds in proteins for SDS-PAGE denaturation, maintenance of reduced cysteine thiols in enzymatic assays, and prevention of protein oxidation during purification.
Principle / Technology DTT reduces disulfide bonds through thiol-disulfide exchange, with the formation of a stable six-membered ring intramolecular disulfide in the oxidized DTT product driving the equilibrium toward complete reduction.
Detection Method Proteins treated with DTT are analyzed by SDS-PAGE, mass spectrometry, or enzymatic activity assays
Sample Type Purified proteins, cell lysates, enzyme preparations, and peptide samples requiring disulfide reduction
