Description An aqueous-based antifade mounting medium containing DAPI (4',6-diamidino-2-phenylindole) nuclear counterstain for the simultaneous mounting and nuclear staining of immunofluorescence-labeled cells and tissue sections.
Intended Use Mounting coverslips onto immunofluorescence-labeled slides while simultaneously staining cell nuclei blue for nuclear localization and cell counting in fluorescence microscopy.
Principle / Technology DAPI binds to AT-rich regions of double-stranded DNA in the minor groove with approximately 20-fold fluorescence enhancement; excitation at 358 nm, emission at 461 nm; compatible with green (FITC), red (Texas Red), and far-red (Cy5) fluorophores.
Detection Method Wide-field and confocal fluorescence microscopy with DAPI filter set (Ex 325–375 nm, Em 435–485 nm)
