Description A ready-to-use 0.5% Crystal Violet solution for the colorimetric quantification of adherent cell biomass. The triarylmethane protein-binding dye stains both viable and fixed cells, providing a measure of total cell density independent of metabolic state and well-suited for endpoint proliferation and cytotoxicity assays.
Intended Use End-point quantification of total adherent cell mass in clonogenic survival assays, cell adhesion studies, and screening for antiproliferative compounds where metabolic-independent measurement of cell number is preferred.
Principle / Technology Crystal Violet (hexamethyl pararosaniline chloride) binds to DNA and protein components of fixed or living cells through ionic and hydrophobic interactions; the bound dye is then extracted with a solubilization solution and measured colorimetrically at 590 nm.
Detection Method Colorimetric absorbance measurement at 590 nm after dye elution using a microplate reader

Reaction Conditions / Protocol Remove culture medium and wash cells gently with PBS; add 0.5% Crystal Violet solution to cover monolayer (50 μL for 96-well, 200 μL for 24-well); incubate 20 minutes at room temperature; wash extensively with water to remove unbound dye; air-dry plate; solubilize with 1% SDS or methanol; measure absorbance at 590 nm
