Description An improved Coomassie dye-based protein assay reagent formulated as a single, ready-to-use solution delivering rapid, linear colorimetric response for protein quantitation. The reagent is optimized for microplate and cuvette spectrophotometric measurement formats.
Intended Use Rapid and accurate total protein concentration determination in cell lysates, tissue homogenates, column chromatography fractions, and purified protein preparations.
Principle / Technology In an acidic environment, Coomassie G-250 dye binds primarily to arginine, histidine, lysine, tyrosine, tryptophan, and phenylalanine residues, shifting the dye's absorbance maximum from 465 nm (brown) to 595 nm (blue).
Detection Method Absorbance measurement at 595 nm using microplate reader or spectrophotometer
Sample Type Protein samples in common biological buffers; compatible with Tris, HEPES, PBS, and other common buffers; incompatible with high concentrations of detergents (>0.1% SDS or Triton X-100)
Performance Range / Specifications Linear range: 100–1500 μg/mL (standard protocol); 1–25 μg/mL (micro protocol with proportionally less reagent)
