Description Cell proliferation and tracing detection reagents are based on CFDA SE for cell proliferation and tracing detection. This reagent consists of CFDA SE powder, solvent, and staining buffer. CFDA SE is a derivative of fluorescein diacetate (FDA) and has cell membrane permeability but does not exhibit fluorescence on its own. When CFDA SE penetrates the cell membrane and enters living cells, it can be catalyzed by esterases in the cytoplasm to form carboxyfluorescein succinimidyl ester (CFSE), which emits strong green fluorescence, cannot penetrate the cell membrane, and remains intact within the cell. CFSE can also spontaneously and irreversibly covalently bind to amino groups within the cell, thereby coupling to cellular proteins. Meanwhile, excess CFDA SE that has not been coupled passively diffuses back into the extracellular culture medium and is removed during subsequent washing steps. Fluorescence in non-dividing cells labeled with CFDA SE is highly stable, with stable labeling lasting for months, making it ideal for cell colony analysis.

Notes 1. Before use, centrifuge the product briefly to the bottom of the tube, then proceed with subsequent experiments. 2. CFDA and SE are easily hydrolyzed and degrade rapidly in aqueous solutions. Avoid contact with water during use. Contact with water during cell labeling is permissible. 3. The CFDA SE solvent may solidify and adhere to the bottom, walls, or lid of the centrifuge tube at low temperatures such as 4°C or in an ice bath. It can be warmed in a water bath at 20–25°C for a short time until completely dissolved before use. 4. This kit has been optimized for the CFDA SE staining system; however, users are advised to gradually determine the optimal working concentration and staining time based on their specific cell type, culture conditions, and application. Different cell types exhibit varying levels of intracellular esterase activity, resulting in differences in staining efficacy. 5. Fluorescent dyes are susceptible to photobleaching. Please avoid exposure to light during handling to minimize fluorescence photobleaching.
