Description CCK-8 (WST-8) is an upgraded alternative to MTT and has significant advantages over MTT or similar products such as XTT and MTS. First, the formate generated by MTT being reduced by certain dehydrogenases in mitochondria is not water-soluble and requires a specific solvent for dissolution; while the formazan produced by WST-8, XTT, and MTS is water-soluble, eliminating the need for subsequent dissolution steps. The formazan produced by WST-8 is more soluble than that produced by XTT and MTS, and WST-8 is more stable than XTT and MTS, resulting in more stable experimental results. Additionally, WST-8 has a broader linear range and higher sensitivity compared to MTT and XTT. Under the presence of an electron-coupling reagent, WST-8 can be reduced by mitochondrial dehydrogenase to form a highly water-soluble orange-yellow formazan product. The intensity of the color is directly proportional to cell proliferation and inversely proportional to cell toxicity.
Application This product can be used for drug screening, cell proliferation assays, cell toxicity assays, tumor drug sensitivity tests, and the detection of bioactive factor activity.

Storage Store at 4°C away from light. For long-term storage, store at -20°C.
Features Good linearity: This product exhibits good linearity at both high and low cell counts. Low toxicity: This product has low toxicity to cells and does not affect subsequent experiments. Ready-to-use: This product is ready to use upon opening and does not require gradient dilution.
Notes 1. Before use, briefly centrifuge the product to the bottom of the tube, then proceed with subsequent experiments. 2. Use a 96-well plate for cell seeding. If the culture time is prolonged, be sure to address evaporation issues. It is recommended to discard the outer ring of the solution and replace it with an equal volume of PBS or culture medium. 3. Before using an enzyme-linked immunosorbent assay (ELISA) reader, ensure there are no bubbles in each well, as they may interfere with the measurement. 4. If OD values are not measured immediately, add 10 μL of 0.1 M HCl solution or 1% w/v SDS solution to each well, and store the culture plate at room temperature in the dark. Absorbance will remain unchanged within 24 hours. 5. If the culture duration is prolonged and the medium color or pH changes, it is recommended to replace the medium with fresh medium before adding the CCK-8 reagent. Culture medium containing phenol red does not affect the measurement of cell activity using this kit. 6. If the sample is a highly turbid cell suspension, it is recommended to set 600 nm (or above 600 nm) as the reference wavelength and subtract the OD value at the reference wavelength. 7. CCK-8 reagent has very low toxicity to cells. It continuously reacts with dehydrogenase in live cells, causing the solution color to darken and the OD value to increase (note: dehydrogenase is continuously produced in live cells). The recommended incubation time is typically 0.5–4 hours.
