Description A highly sensitive, single-solution cell viability assay using the water-soluble tetrazolium salt WST-8. The WST-8 reagent produces a soluble orange formazan dye upon reduction by cellular dehydrogenases, eliminating the need for organic solvent dissolution steps required by MTT.
Intended Use High-throughput cell proliferation and cytotoxicity screening in drug discovery, toxicology studies, and cell biology research using mammalian cell cultures.
Principle / Technology WST-8 (2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium) is reduced by NADH/NADPH-dependent dehydrogenases in viable cells to a water-soluble formazan dye with absorbance at 450 nm; an intermediate electron carrier (1-methoxy PMS) enhances the reaction rate.
Detection Method Colorimetric absorbance measurement at 450 nm using a microplate reader with reference wavelength at 600–650 nm
